Enhancing CAR-T Cell Function with VHHs: Overcoming Exhaustion and Improving Targeting

It is know that T-cells used in CAR-T at times become exhausted prematurely before performing their actual therapeutic role. This may be caused by VL and VH mispairing (originating from the scFvs), triggering premature signal cascade activation. The monomeric nature of VHHs, in addition to their increased hydrophilicity compared to scFvs, points to VHHs as a potent instrument to direct T-cells to home in to the tumor.
